dc.description.abstract There are over 30 species of terrestrial orchids in Indiana, all of which are threatened or endangered because of man's encroachment into their natural habitats. In order to protect our native orchids and identify endangered populations, we have assessed the genetic variation within one species, Aplectru n hyemale. Twenty samples were collected from three different forests. The samples collected were compared for both within and between population genetic variability. Various DNA isolation protocols were compared, the polymerase chain reaction (PCR) conditions optimized, and five Random Amplified Polymorphic DNA (RAPD) primers tested.
